A new study on endangered Pacific Northwest killer whales?is getting noticed not just for its findings -- less salmon to eat is a much bigger stress factor than whale watching boats -- but for who helped out: namely, a black Labrador trained to detect killer whale scat.?

Cited as "the dog" in the peer-reviewed study, Tucker is credited with allowing researchers to get a better sampling of the resident killer whales in waters off Washington state and Canada's British Columbia while monitoring from a distance.

"We debated about naming Tucker," lead researcher Katherine Ayres told msnbc.com, "but decided it was more 'scientific' to say 'the dog'.

"We also wanted this to be a general method that could be applied with other dogs in the future so we didn't want to make it too specific," she added. "However, you will see that he has a big -- and much deserved -- shout out in the acknowledgements."

Indeed, after thanking numerous humans, the authors concluded with: "Special thanks go to Tucker, the Conservation Canine, for his keen nose and assistance with non-invasive fecal sampling."

"Use of a detection dog enabled us to sample at an average distance of 400 meters from the target whale(s), minimizing any potential disturbance from the research vessel," the researchers wrote in the study published Wednesday in PLoS One.

Prior to Tucker, researchers had to closely follow orca pods to find and collect scat samples. But that probably led to shy orcas staying away from research boats,?Ayres said, making for a less representative sampling of the population.

With Tucker, "we were not influenced to follow whales that are naturally charismatic" toward humans, such as large males or mothers with calves, said Ayres, a?pet-behavior consultant who led the research while a University of Washington doctoral student in biology.

The scat was used to detect hormones released by "southern resident killer whales," a population listed as endangered due to low numbers in recent years. Levels of thyroid hormones, which slowly regulate metabolism based on food availability, showed the orcas were best fed when first arriving in the Salish Sea off the coast of Washington and British Columbia.

The researchers also found that fast-acting stress hormones known as glucocorticoids, which are triggered by immediate danger and food stress, did not spike when whale watching boats reached their annual peak around the orcas.

"As Chinook salmon increased, stress levels went down," said Ayres.

The study did, however, find a cumulative impact from vessels on stress. "Say the only place in town to eat is a noisy/crowded bar," Ayres said by way of analogy. "If that bar has all the food you like and plenty of it, you might not be that bothered by the noise. However, if you are starving and the buffet only has rice and potatoes, you might start to notice the noisiness more and you might become more stressed by it."

"Fish matter most to the southern resident killer whales," added study co-author Sam Wasser, director of the?University of Washington's Center for Conservation Biology. "Even if boats are important to consider, the way you minimize that impact is to keep the fish levels high," he said in a statement issued with the study.

The researchers wrote that Tucker was chosen "for its obsessive drive to play with a ball."?In training, when he located a sample, he was praised with a short play session.

Out on the water, they added,?"the dog indicated sample detection by changing his behavior from a relaxed sit or stand to leaning over the bow of the vessel with tensed muscles, anticipating a reward." Tucker maintained that behavior as the scent intensified, and alerted the handler when the concentration was weakening.

"As we got close to the scat, the dog often stood up and began to whimper, presumably because the scent was surrounding the vessel and he could no longer follow a concentration gradient."

The technique originated with Wasser, who has applied dogs to similar studies with other species over the last decade, Ayres said.

Tucker was first deployed?in 2008, and?the study cements his legacy as a pioneer. "This is the first study using scat-detection dogs to locate killer whale feces," Ayres said.?

Nowadays, Tucker is still used for orca studies but he's also expanded his range.

Gigs have included working on St. Lucia in the Caribbean to track iguanas. "He also was trained on moose/caribou/wolf in Alberta, Canada, in the winter time when we weren't using him for whales in the summertime."?Ayres said.

"That way," she added, he has "a job almost?year round."

Keeping up with embryogenesis: New microscope tracks cells as they move and divide

ScienceDaily (June 5, 2012) ? The transformation of a fertilized egg into a functioning animal requires thousands of cell divisions and intricate rearrangements of those cells. That process is captured with unprecedented speed and precision by a new imaging technology, developed at the Howard Hughes Medical Institute's Janelia Farm Research Campus, which lets users track each cell in an embryo as it takes shape over hours or days.

In a single movie -- a compilation of a million images captured over about 20 hours -- viewers can see biological structures begin to emerge as a simple cluster of cells morphs into an elongated body with tens of thousands of densely packed cells. The movie concludes when the embryo begins to twitch, driven by contractions of its newly formed muscles -- moments before the hatching of a fruit fly larva just half a millimeter long.

The imaging technology -- driven by a high-speed, non-invasive light microscope that simultaneously captures images from four different angles -- is described in the June 3, 2012, issue of the journal Nature Methods. According to its inventor, Janelia Farm fellow Philipp Keller, the new technology will enable quantitative analyses of developmental processes that were not possible with previous imaging methods. "This is not just a quantitative improvement [over previous technologies]," Keller says. "It is the difference between being able to quantitatively measure this process and not even being able to follow the cells."

Like many developmental biologists, Keller, who works at the interface of biology, physics, and computer science, wants to understand how a single cell transforms into a complex organism. "It's a really complex process, and if you want to understand it, I think it is really important that you are able to visualize and observe it," he says. But conventional light microscopes are too slow to follow the rapid cellular migrations that occur at this stage of life, and also tend to damage a living embryo with light before its development is complete. So scientists have been forced to piece together snapshots of different stages of development from multiple organisms, and make assumptions about the changes that must have occurred in between what they actually see.

"This approach is incompatible with the highly dynamic process of development," Keller says. "Snapshots just don't capture the essence of it." Furthermore, cells may divide and rearrange themselves differently each time development occurs, so combining observations from multiple organisms could be misleading.

To better follow development, Keller says, scientists needed a way to image at high speeds, over long periods of time, without interfering with the processes they wanted to observe. Frustrated by the limitations of existing technology, Keller set out to design a microscope that would let him investigate the biological questions that fascinated him most.

The new microscope, which Keller's team calls the SiMView light sheet microscope, builds on technology Keller developed with colleagues at the European Molecular Biology Laboratory in Heidelberg, Germany to address one of the concerns associated with conventional light microscopy: the damaging effects of exposing a sample to light. Light is essential to activate and visualize the fluorescent labels researchers attach to molecules inside cells, but too much of it damages cells and causes fluorescence to fade. Keller and his colleagues reduced this damage by illuminating only a thin section of a sample at a time with a scanned sheet of laser light, while a detector records the part of the sample being lit up.

The light sheet technique was fast, and could capture the rapid process of development in samples that were not too thick or opaque. In larger or less translucent embryos, however, light from the microscope would scatter as it passed through layers of cells, blurring the image. In a fruit fly embryo, for example, only about 30 percent would be visible.

"You really lose a lot of the information," Keller says. "If you want to study cellular dynamics during development to get a complete, dynamic building plan of the entire animal, you can't afford to not see parts of the sample."

The solution was, in concept, simple: build a light sheet microscope that collects images from multiple views at the same time. "Instead of looking at the sample from one direction, we look at it from four directions at the same time," Keller explains.

In the new microscope, two light sheets illuminate a sample from opposite directions, and two detectors collect the resulting fluorescence. Each combination of illumination and detection lens provides a different perspective.

Capturing those four views simultaneously eliminates delays caused by rotating a sample repeatedly into new positions -- a traditional approach to multi-view imaging -- and makes the microscope very fast. "We tried to push the speed as much as we could," Keller says. This version is as much as 50 times faster than light sheet microscopes that capture multiple views sequentially, he says.

Coordinating the microscope's components to illuminate the sample and detect fluorescence at the right time and place requires tremendous precision: even fluctuations in room temperature during the course of an experiment can shift components out of alignment. Timing is equally important: light sheets that cross paths can blur an image, so they must be staggered by a few milliseconds. To maintain precision, the SiMView is equipped with an electronics system that makes adjustments in real time.

Once Raju Tomer, a researcher in the lab, and Keller had implemented the design of the microscope, the team had to confront another hurdle: managing and interpreting the tremendous amount of data it generated. The default setting for the microscope, Keller says, is to collect 350 megabytes of data per second. Imaging a single sample over a full day can produce up to several tens of terabytes of data -- about the amount of data stored in the complete Library of Congress archive. For experiments designed to compare development under different genetics or conditions, many times that amount will need to be stored and analyzed. "We want the data," Keller says. "It's not a problem, but it is a challenge."

Anticipating that challenge, lab members Fernando Amat and Khaled Khairy devoted their attention to developing computational methods to identify and track individual cells in the movies produced by the microscope. Those methods are an essential part of what Keller calls a complete technology framework for imaging living samples. The team is now collaborating with Janelia Farm colleagues Kristin Branson and Eugene Myers to develop new computational strategies to automate data analysis,

Having focused on developing that technology for the past two years, Keller says he is looking forward to shifting more of his attention to biology. He anticipates that his lab will continue adapting the microscope and making its computational processing more efficient and powerful. "But we now have the technology to answer most of the questions we are interested in," he says.

Included in the Nature Methods publication is a movie showing the complete development of the nervous system in a fruit fly embryo. Keller explains that individual cells can be followed as they give rise to sensory organs, brain lobes, and other structures. At the same time, the microscope offers resolution high enough to zoom in and see the fast dynamic processes at the tip of a neuron's axon.

Anticipating future experiments, Keller says the microscope will let his team not only trace cell lineages in the embryo, but begin to manipulate development to probe the mechanisms that control it. With Janelia colleagues, he has begun to explore other possibilities, tracking development in larger, more complex samples, such as mouse embryos and fruit fly larvae, and monitoring signaling in the zebrafish brain.

"The microscope isn't the limiting factor anymore," Keller says. "That's really exciting."

'Harry Potter': An MTV Movie Awards History

Following Best Cast and Best Hero wins for final film, we look back at our favorite Movie Awards moments with the Boy Who Lived.

UNIVERSAL CITY, California — With its Best Cast and Best Hero wins at the 2012 MTV Movie Awards, the "Harry Potter" era in MTV history has come to a close.

MTV has been a fan of "Harry Potter" since the beginning. Literally. It's bittersweet to have to celebrate the series for the last time with the big wins for "Harry Potter and the Deathly Hallows: Part 2," so we decided to take a look back at our favorite Movie Awards memories with The Boy Who Lived.

The series didn't earn its first Movie Award nominations until 2006, for "Harry Potter and the Goblet of Fire." It got nods for Best Hero (Daniel Radcliffe), Best On-Screen Team (Radcliffe, Emma Watson and Rupert Grint) and Best Villain (Ralph Fiennes), but didn't take home any awards. "Potter" struck out yet again with "Harry Potter and the Order of the Phoenix" when it lost both Best Summer Movie You Haven't Seen Yet in 2007 and Best Kiss (between Daniel Radcliffe and Katie Leung) in 2008.

There was a year of downtime between "Potter" nominations because the series didn't have a film come out in 2009, but that doesn't mean it wasn't represented at the Movie Awards. Radcliffe, Grint and Watson presented a "Harry Potter and the Half-Blood Prince" clip showing Dumbledore using the Pensieve to show Harry the first time he met Tom Riddle, who later would become Lord Voldemort.

It wasn't until 2010, when "Half-Blood Prince" was up for four awards, that the series took home its first Golden Popcorn. The film lost for Best Female (Watson), Best Male (Radcliffe) and Best Movie, but Tom Felton took home the award for Best Villain. That ended up being one of the most memorable acceptance speeches in recent Movie Awards history as Mark Wahlberg kept interrupting the young Brit while he was trying to talk.

Fans were duly outraged, but Wahlberg was quick to apologize to Felton backstage — and we caught it on tape! "We were only kidding," Wahlberg said, patting Felton on the back. "Sorry about that, great job. ... Thanks for being a good sport."

That year also marked the second time the Movie Awards showed a sneak peek of an upcoming "Potter" movie. We premiered a teaser trailer for both parts of "Deathly Hallows," which ended up being pretty freaking awesome.

Felton took home the series' second Golden Popcorn the next year (again for Best Villain), though "Harry Potter and the Deathly Hallows, Part 1" lost for Best Female, Best Kiss (Watson and Radcliffe), Best Male, Best Movie and Best Fight. That night was another memorable one for Felton, as Foo Fighters leading man Dave Grohl congratulated him on camera following his win.

One of the most talked about moments of the 2011 MTV Movie Awards was the premiere of the forest scene from "Harry Potter and the Deathly Hallows, Part 2." It was one of the more chilling moments from the movie, and one that boded well for the final installment of the "Harry Potter" saga.

Fans were ultimately disappointed with the fact that "Potter" only took home one award in 2011, especially since it lost most of the Golden Popcorns to "The Twilight Saga: Eclipse." With the voting rules changed this year, that means that the competition in 2012 was less about a battle of the fandoms and more about the best movie winning.

In the end, "Harry Potter and the Deathly Hallows, Part 2" didn't win every award it was up for this year. It lost for Best Fight, Best Female, Best Male, Best Kiss (Watson and Grint) and Best Movie, but at least won for Best Cast and Best Hero. At the end of the day, "Harry Potter" really does deserve both of those awards.

Fans could argue for hours over whether "Harry Potter" or "The Hunger Games" deserve the awards they won, but it seems fair to say that "Harry Potter" has consistently had one of the best ensemble casts in recent memory. It's not just Radcliffe, Watson, Felton and Grint who won the award, but everyone who participated in the series' eight films. As for the Best Hero win, considering that Harry defeated one of most evil beings of all time (you know, He Who Must Not Be Named), that's quite a heroic feat. Those seem like suitable thank-yous to the "Potter" crew for making one of the most beloved film franchises in movie history.

The recall battle has raged for months, sparked by Walker's elimination last year of most collective bargaining rights for public-sector unions. That led to huge street protests in Madison, the state capital, and to the recall effort that will culminate in Tuesday's vote.

Walker's national supporters see the recall as a referendum on his take-no-prisoners brand of fiscal conservatism, and both sides in the presidential race plan to scour Tuesday's results to gauge the mood of the electorate heading into November's bigger battles.

"The vote here will definitely be seen as a harbinger of things to come," said Wisconsin-based Republican consultant Mark Graul. "The winning side will have tremendous momentum, and there will be implications far beyond Wisconsin."

Wisconsin has not backed a Republican in a presidential election since it went for Ronald Reagan in 1984, but Republicans hope a recall win will demoralize Democrats and sharply boost Romney's hopes.

Most recent polls have shown Walker ahead of Barrett, who is the mayor of Milwaukee, by about 7 percentage points in the recall election. But in a reflection of how difficult it can be to read the political tea leaves in a "swing" state such as Wisconsin, polls have shown Obama ahead of Romney by 6 to 10 points.

Obama easily captured Wisconsin by 14 percentage points in the 2008 election, when he defeated Republican John McCain. Two years later, Republicans in Wisconsin roared back to elect Walker, defeat Democratic U.S. Senator Russ Feingold and take over the state legislature.

"If the governor is successful" in warding off a recall, "it will show the mood of voters is closer to where it was in 2010 than in 2008," Graul said. "And that could portend trouble for Obama here and in other similar states, like Ohio," another Midwestern state that will be perhaps the most significant battleground in the presidential race.

Walker's bulldozing approach to cutting state budgets has made him what Barrett derisively calls "a rock star for the far right."

Conservative groups such as Americans for Prosperity, funded by the billionaire conservative brothers Charles and David Koch, have poured millions of dollars into Wisconsin and plastered the state's airwaves with anti-Barrett ads. It is part of an effort to make a statement for their conservative cause - and set a tone for the presidential election.

Walker has raised $31 million to Barrett's $4.2 million, and by late April about two-thirds of Walker's 2012 donations had come from out-of-state donors, according to finance reports compiled by the Wisconsin Democracy Campaign, which tracks money in state politics.

"If Governor Walker survives this, then national Republican and conservative leaders are going to see him as the spearhead for a movement," said Joe Heim, a political scientist at the University of Wisconsin-La Crosse.

Romney and Obama have avoided much involvement in the Wisconsin race, perhaps a sign that the presidential candidates are not willing to risk being tied too closely with either side.

When Romney was in Wisconsin to campaign in the state's April 3 presidential primary he called Walker a "hero." Obama endorsed Barrett in a brief statement after the May 8 Democratic primary but has said nothing publicly.

Even some Democrats think Barrett has run an ineffective, uninspiring campaign with his attack on the governor's leadership that moved beyond Walker's push for anti-union legislation.

Barrett has targeted Walker's record on creating jobs and raised questions about a criminal probe into whether Walker's aides when he was Milwaukee County executive did political fundraising on public time.

But after months of polarized battle over Walker's agenda, most Wisconsin voters seem to have made up their minds. A recent Marquette University law school poll said just 3 percent of voters remain undecided; a Democratic poll this week put the figure at 1 percent.

That makes voter turnout a potentially huge factor in the recall vote. Both sides have promised unprecedented get-out-the-vote operations.

"We've said all along this will be very close and will be decided by turnout, and we think we'll have the most effective turnout operation," Wisconsin Democratic Party Chairman Mike Tate said.

While Democrats have a slight edge in voter registration in Wisconsin, Republicans seem to have at least a slight edge in enthusiasm. The Marquette poll found 91 percent of Republicans were "absolutely certain" to vote, compared with 83 percent of both Democrats and independents.

'A DRY RUN'

"When I make calls, I get Republicans who are really angry this is happening. The Democrats are mad too, but I don't think they are as passionate about it," said Chris Johnson, a retired teacher who volunteers at a Walker field office in La Crosse.

Walker has opened 22 offices around the state to direct his turnout operations, and he said volunteers have made more than 2 million calls to voters - more than during his entire 2010 race.

Volunteers made more than 200,000 contacts with voters on a recent weekend day, and other Midwestern state Republican parties are setting up their own phone banks for Walker.

Volunteers with the state Democratic Party knocked on 100,000 doors that same weekend day, and labor groups also have set up their own networks of more than two dozen field offices.

The turnout operations will serve as a test run for November's presidential contest. Tate said there was significant overlap, for example, between the state Democrats' field staff and Obama's campaign.

Some Wisconsin Democrats have grumbled about the lack of support from the national party.

The Republican Governors Association was one of the biggest spenders in the race, pumping in nearly $8 million, while the Democratic National Committee only sent out a late fund-raising appeal last week.

State officials are predicting a big turnout on Tuesday, and Democrats are targeting people who voted in 2008 but not 2010. They are hoping to get 150,000 to 200,000 recall voters to the polls who did not cast a ballot in 2010 - when Walker won by about 100,000 votes.

The risk is that the losers will be demoralized after the intensity of the recall fight, making it harder to get them back to work in the campaigns for November's presidential election.

"It's very easy to burn out the average political volunteer. I'm not sure how easy it will be to get people back through the door in September," said Brian Westrate, the Republican chairman in Wisconsin's Eau Claire County. "But it will be a lot easier if we win."
